Activation of plasminogen by trypsin and plasmin.

چکیده
teolytic enzyme plasmin (fibrinolysin) is supposed to be present in normal blood plasma in an inactive form, called plasminogen (proplasmin). Anxiety, strenuous exercise, hemorrhage, burn , t rauma (‘ ‘tourniquet shock’ ‘) and epinephrine admmistration have heels found to produce increased fibrinolytic activity (plasmin formation) in the blood. 3 The physiologic mechanism of activation of proplasmin in the normal organism is at present unknown. Since plasmin is al)le to digest fibrinogen and prothrombin,2 and to lyse fibrin clots, its presence in blood may lead to fatal failure of the hemostatic mechanism. Human plasminogen may be converted to plasmin by treatment with chloroform4 or with streptokinase5 or staphylokinase.6 Bovine plasminogen, however, is apparently resistant to the action of these kinases and the only known w’ay to bring about conversion to the active enzyme is by treatment with chloroform. However, it is apparent that none of the above mentioned activators can he

Background: Plasminogen has a central role in fibrinolyrtic system can activate through various activators (PAs) to its active form plasmin and perfoem its vital function that is fibrin clot lysis. Furthermore the fibrinolyrtic system plays a major role in angiogenesis. The fibrinolyrtic system activation control cell migration and invasion. In addition to this, plasmin regulates tumor growth. ...

Background: Human plasminogen is a plasma glycoprotein synthesized mainly in the liver. Conversion of plasminogen to plasmin by plasminogen activators is a key event in the fibrinolytic system. In this study, we investigated the effects of two anti-human plasminogen monoclonal antibodies, A1D12 and MC2B8 on Glu-plasminogen activation in presence of u-PA, t-PA and streptokinase. Methods: Produci...

Human and animal sera contain a globulin, plasminogen, which in the presence of activators is rapidly converted to plasmin, a proteolytic enzyme active at neutral hydrogen ion concentrations. Activators specific for plasminogen include streptokinase and staphylokinase of bacterial origin, urokinase and plasma activator found in body fluids, and fibrinokinase derived from tissues. Trypsin, an ac...

متن کاملPreparation of Plasminogen by Affinity Chromatography
Background: Plasminogen is one of the compounds derived from human plasma. Activation of plasminogen produces plasmin. Plasmin is able to lyse fibrinogen, fibrin, and some other human plasma proteins. The aim of the present work was to study the separation of human plasminogen by affinity chromatography using gel lysine Sepharose. Materials and Methods: Normal human plasma was used as the...
متن کاملThe effects of fibrinogen and its cleavage products on the kinetics of plasminogen activation by urokinase and subsequent plasmin activity.
The effects of fibrinogen and its plasmic cleavage fragments on the activation of Glu-, Lys-, and Val442- plasminogen by urokinase were investigated. A possible explanation for the large variations in the published steady state parameters for Glu-plasminogen activation is the undetected formation of Lys-plasminogen and its subsequent more rapid activation to plasmin. When Lys-plasminogen format...
